线粒体证据揭示秦陵修陵劳工的来源

200433;2.吉林大学 边疆考古研究中心 古DNA实验室,长春 130012;3.中国社会科学院 考古研究所,北京 100710;4.陕西省文物与考古研究所,西安 710054;5.中国科学院 上海生命科学研究院-马普学会 计算生物学伙伴研究所,上海 200031.摘要:越来越多的研究试图利用古DNA的手段揭示古代人类个体的来源。2003年秦始皇陵秦俑坑附近出土的一批人骨被推测是建陵劳工的遗骸,年代大约在2200年前。我们通过克隆测序和PCR-PRLP的方法,成功地从19个骨样本中得到线粒体HVR区和编码区的SNP位点的序列信息。为了揭示这些劳工的来源,我们将所得序列与中国现代人的32个群体(2164个体)的线粒体序列分别在个体和群体水平上作比较,结果表明秦陵劳工墓的劳工来源是多样的。本文详尽地介绍了古DNA的操作方法、序列信息的甄别和数据分析方法。
